As a child Rappaccini was a brilliant geneticist, botanist, and inventor. During a science fair, she was planning to reveal her instantly regenerating fruit tree. During this event versions of M.O.D.O.K. and Jodie from an alternate future approached her about building a time machine, a thing she was currently unable to do. Unfortunately M.O.D.O.K. destroyed her project as a test of his appetite, causing a school guidance counsellor to cruelly dismiss her efforts at science and place her on the path of supervillainy.
Working for A.I.M.
In 2009, Monica interviewed for a job at A.I.M. to work with her idol, M.O.D.O.K. She was hired as a Science Drone and on her first day she killed a high-profile Avenger, but M.O.D.O.K. took the credit.
Over the years, Monica was promoted from Science Drone to one of the lead scientist in charge of R&D. Never taking a single vacation day in her 10 years with A.I.M.; Monica created a time machine, developed a black hole gun, genetically engineered creatures, and began working on manatee soldiers. At some point, Monica cloned a male version of herself to impregnate her and she gave birth to her daughter Carmilla.
At one point, Jodie, M.O.D.O.K.'s estranged wife, broke in to A.I.M. in an attempt to support the debunked narrative of her son having a peanut allergy. After discussing M.O.D.O.K. with her, Monica implanted Lou with a peanut allergy to help support Jodie's narrative 
One day, while experimenting on Manatees, Monica was told to use her vacation days by Austin, the GRUMBL appointed head of A.I.M. to start using her vacation days. M.O.D.O.K. confided in her, as he was currently attempting to kill Iron Man with a black hole gun and was being stifled by A.I.M's bureaucracy. Their similar frustrations resulting in them banding together, with M.O.D.O.K. and Monica using a Silicone-Based humanoid body-doubles to pose as them for a mandated seminar while they worked to access the computer of Doris, a key figure in A.I.M. decision making. Distracting Doris by interfering with her connection, M.O.D.O.K. and Monica used Gary's birthday to draw Daphne, Doris' assistant, away from her office. Hacking into Doris' computer, they were able to rescind all of Monica's vacation days and grant M.O.D.O.K. access to the black hole gun. Monica then betrayed M.O.D.O.K. as revenge for his poor treatment of her, revealing his plans to Austin and blaming the plan entirely on him. M.O.D.O.K. and Monica engaged in a fight, resulting in Monica being crowned as the Scientist Supreme. However, GRUMBL then used this as an opportunity to seize most of Monica's equipment and assign her on the Jiggle-Os. The only item she was able to keep was the skull of the Avenger she killed. 
Upon M.O.D.O.K. doing his job as Mailman, Monica became paranoid about a wider scheme of M.O.D.O.K. to kill her, resorting to measures such as having her daughter shrink with Pym Particles and go inside her body and kidnapping Gary. Eventually, M.O.D.O.K. revealed his plan to be that of A-I-M 2, a legally distinct company where he hoped to have Monica as scientist supreme. Disappointed with the corporate culture of GRUMBL, Monica agreed to this proposal, selling her shares to Iron Man and giving him a controlling stake in the company. 
Monica is brillant, maniacal, cunning, suspicious, and prone to anger.
