The year is 1933, and a squad of police burst into the Daily Bugle after getting a call from someone claiming they were J Jonah Jameson who said they were dying. Throwing open the door to Jameson's office, they find Jameson bleeding in his chair with Spider-Man crouched over him, a gun in his hand. The police open fire at the wall-crawler, and Spider-Man jumps through an open and into the night. None of the bullets find their mark, but he does get nicked by one of them.

Time jumps backwards to J Jonah Jameson telling his reporter, Ben Urich, to go out into the streets and get pictures of the poverty that is occuring in New York. Ben goes to a section of the city that has been overrun by homeless shanties, and in the middle of those shanties he finds the famed May Parker giving a speech to a small crowd of people. May is railing against the current presidents and political parties of the United States of America and is demanding people to take action and support socialism instead. A group of men approach, which Ben identifies as the Enforcers, and the three gang members tell May to stop spreading her political message. When May resists, the Enforcers begin to rough her up as well as her nephew, Peter Parker, who tries to step in and defend his aunt. Ben Urich stops the Enforcers by taking a picture of them and threatening to put it in the paper, and the Enforcers are forced to comply because Urich is "untouchable" to them.

Urich drives Peter and May Parker back to their home where they run a soup kitchen to give homeless people free meals and shelter. Peter is furious at the way the world works, so to give him a taste of reality and to damper his hopes of making a difference, Urich takes Peter to a speakeasy run by Felicia Hardy called the Black Cat where all of the upper echelon of the city frequent. During their trip to the speakeasy, Peter's anger is set off by an encounter with Norman Osborn, who people call "the Goblin", and who is the man responsible for the death of Peter's Uncle Ben.

Seeing that Peter was not subdued by the corruption they witnessed in the speakeasy, Urich takes Peter under his wing and gets him a job at the Daily Bugle as Urich's photograph assistant. Together they roam the city taking pictures of awful injustices and people living in horrible conditions because of the economic depression. Witnessing each of the injustices, however, just continues to make Peter more angry instead of breaking his spirit. Finally one night Peter opens up to Urich about his life and about how his uncle was killed by the Goblin's men when they let out wild dogs on him.

Ben Urich remembers to himself the night that Peter's uncle was killed, and remembers how his body was not torn apart by dogs but rather by one of the Goblin's Enforcers - the cannibalistic Vulture. Ben also reveals that the reason he can remember the night is because he himself was there to watch it at the time it happened because he was in league with the Goblin's men.

Peter Parker and Ben Urich are developing photos from a recent apartment building fire when Peter notices one of Norman Osborn (the Goblin)'s henchmen in one of the shots, linking the Goblin to the instigation fire. Urich brushes it off and responds with a snarky comment when Peter tries to tell him about the ideals his deceased Uncle Ben instilled in him.

Later, Peter arrives at Ben Urich's home to give him news about their photograph in the paper, only to discover Urich zoned out on a heroin drug high. While Peter is inspecting Urich's drug needles, the phone rings and Peter picks it up to hear someone giving a tip about an upcoming photo opportunity of the Goblin's men unloading ancient artifacts at the Pier at 1 AM.

Peter goes to the Pier at the time of the unloading and hides in the rafters. He watches as the Goblin's henchmen accidentally drop a case they are unloading, from which swarms of spiders come crawling out and bite one of the Goblin's Enforcers to death. Peter then notices one of the spiders descend on a strand of webbing near his face, but he tries his hardest not to scream even as the spider bites him. Peter goes into a vision where he sees a giant demonic spider tell him that its bite is only fatal to those with evil intent, and then Peter blacks out. When he wakes up, he is wrapped in a web cocoon hanging from the rafters, and he realizes that he has enhanced agility abilities.

The next day in Osborn's headquarters, Ben Urich arrives to sell the Goblin the picture of his henchman at the fire in exchange for the picture not being released to the newspapers. Goblin pays Urich for the blackmail just as Peter crashes through the window wearing a small mask around his face. Peter takes out the Goblin's bodyguards before threatening the crime lord himself, but Osborn is unafraid and tells Peter that he will have to kill him to do any sort of damage to his empire. Just then Peter notices the photo from the fire that he helped develop days before, and turns to see Urich escaping out the doors. Leaving the Goblin, Peter chases Urich out into the street and yells at him, appalled at his unethical actions, and tells Urich that he never wants to see him again. Urich recognizes Peter, but before any other interaction can happen the Goblin and his men come out of the building and begin firing at Peter. Peter escapes over a fence, and Urich, now disgusted at his own actions, throws the Goblin's money back at the crime boss and lets him know that Urich is done dealing with the Goblin. The Goblin, realizing that Urich no longer has anything to lose to keep him quiet, arranges for measures to be taken to stop any leaks.

Urich returns home and calls J Jonah Jameson to tell him that he has pictures linking Norman Osborn to numerous crimes. Jameson tells Urich to stay at his home for his own safety, and that he will come over himself to pick up the pictures. When Jameson arrives at Urich's home, however, Urich opens the door only to have Jameson shoot him in the head, killing him. Jameson then calls the Goblin to let him know Urich has been eliminated.

Meanwhile in his own home, Peter finishes sewing together his Uncle Ben's old pilot uniform into a costume. Peter dons the costume and christens himself Spider-Man, swearing to take revenge on Osborn.

Peter decides that he needs Ben Urich's help to stop the Goblin, so he goes to Urich's home. However, upon arriving he finds Urich dead[1] and calls it in to the police. Later, as Peter is walking the town, he is invited into the Black Cat speakeasy to meet with Felicia Hardy, the bar's owner. Felicia reveals to Peter that his inspiration to Urich to do the right thing is what got Urich killed, and she gives Peter all of Urich's files with incriminating evidence on the Goblin (Norman Osborn) and his men. In his headquarters, the Goblin himself is furious and increases the intensity of the search to find whoever has the files on his operation.

After being advised by Felicia not to give the files to the Daily Bugle, Peter begins using the information in the files to attack the Goblin's operations as Spider-Man. After a number of nights of this, other crime bosses begin to lose faith in the Goblin's protection of them and give him an ultimatum to eliminate Spider-Man. This motivates the Goblin to search even harder for the person tipping off Spider-Man with information about Osborn's operations, and while he is brooding his Enforcer Ox mentions seeing Peter going to meet Felicia Hardy after Urich died. The Goblin decides to kidnap both Peter and Felicia to figure out what they know and where the information on the Goblin's operations is coming from.

Later, Peter is reading the Daily Bugle's smear campaigns about Spider-Man and decides it is time to pay Jameson a visit. He enters Jameson's office only to find him shot and nearly dead in his chair. Jameson mutters something about "the master of disguise" before breathing his last, and just then police burst into the room.[2] Spider-Man escapes through a window, getting grazed by a bullet, and after he has successfully escaped realizes how dangerous his life has become. Peter can't understand why Jameson was shot and goes looking through Urich's files to find answers. After searching through the files on Osborn's Enforcers, he finally stumbles upon something that tips him off.

Spider-Man heads over to the morgue and finds the dead body of Jameson. He presses into the face of Jameson to find that in fact it was not Jameson at all who died, but rather an ex-Enforcer of Osborn's, the Chameleon. This leaves Peter wondering where the real Jameson is. In another part of town, the answer to Peter's question is revealed as Jameson is sitting in a cage, trapped by Osborn. The Goblin brings a gagged Felicia Hardy to join Jameson in the cage, and mentions that he is still waiting for their final guest, Peter Parker, to arrive.

In Peter Parker's home, Aunt May walks into his room looking for the boy. Unbeknownst to her, the Vulture is in the room, looming directly overhead as she walks inside.

Spider-Man is shocked to find a lobotomized Robbie Robertson, and plans to help the other captured black prisoners to escape.

Doctor Octavius is very angered to hear from Crime Master that he and his benefactors of the Friends of New Germany are abandoning his entire research and plans to destroy everything, which includes killing the prisoners, from being caught by the authorities. After the Crime Master forces Octavius to contact Joseph Ansell through telephone, Ansell confirms to him that his experiments are needed to end. But, Ansell privately tells Octavius that he can eliminate Crime Master for being a liability after everything is dealt with.

Crime Master then orders his men to kill the prisoners. Fortunately, Spider-Man attacks and takes their guns away, leaving them at the hands of the escaped prisoners. Crime Master quickly spots Robbie and held him hostage, and demands Spider-Man to back off, while offhandedly stating that he killed Felicia Hardy. Then one of Crime Master's men urgently informs his boss that FBI agents have arrived and are already raiding the jetty. Crime Master orders his men to hold off the agents, but Spider-Man tells him and his goons that it is pointless as anyone who shoots back will be serving time on death-row with their boss, given to his atrocities on Ellis Island. Spider-Man's threats work, as Crime Master's men flee in panic and abandoning their boss. As Crime Master still holds Robbie, Dr. Octavius arrives and informs Crime Master that there is nowhere for him to escape even from Spider-Man and suddenly attacks him with his scalpel arms, leaving Robbie to escape from harm. After Spider-Man had Robbie escape with the other prisoners, Octavius impales Crime Master and mercilessly eviscerate him, as the dying crime boss begs for help from Spider-Man, who only coldly watches on.

After killing Crime Master, Octavius offer his hope to Spider-Man that killing the crime boss would offered good word in the end on his trial if captured. But Spider-Man is unsway and, still angry for what Octavius had done to Robbie, violently subdues the doctor. Just as Spider-Man is about to strangle Octavius, Agent Jean De Wolfe arrives and stop him. De Wolfe then reveals to Spider-Man that Felicia Hardy, who is reveal to be alive, called him about the island's experiments which raises Spider-Man's hope.

Spider-Man and everyone leave Ellis Island, with all culprits connecting to Octavius' experiments captured. Upon arriving on Manhattan, Spider-Man leaves to see Felicia. Arriving At Felicia's apartment, Spider-Man finds her bodyguard Lippy, who informs him that Felicia is being taken care in a private clinic and had decided not to see Spider-Man ever again because it was his fault that caused her condition.

A month later, Peter, Aunt May and Mary Jane Watson visit the Robertson's residence where Robbie is being taken care by his family. Robbie's physical condition is fine, but his brain damage couldn't be medically repair. Carver informs Peter that Agent De Wolfe had called and informed him that Dr. Octavius cannot be held at trial as he had previously worked on government projects that would publicly expose state secrets, and is instead being deported to Germany. Peter is sicken to hear this and regretfully apologizes to Robbie. Mary Jane take Peter outside and talk him through from blaming himself for losing the people he loves, and assures that she will always be with him.

Meanwhile, Dr. Octavius arrives in Berlin and being awaited by German officials lead by Heinrich Himmler. Just as Octavius meets Himmler, the man then notice the doctor's congenital disability and contemptibly insults him, stating that Octavius is "useless" in accordance to the Nazis' eugenicist ideology and refuses to see otherwise. Himmler then coldly leaves and showing no support for Octavius, as the doctor is left alone and deeply dejected.

In the end, Spider-Man narrates that Felicia has recover but hasn't leave her apartment and never had any visitor apart from Lippy. Though Felecia stated that she never wanted to see Spider-Man, he has been respecting her privacy and had been watching her on her balcony, while looking out over the city. Lippy had told Spider-Man that after Felicia had called De Wolfe, she had never told him nor anyone about her injuries, which left her walking with a stick. At the last page, Felicia is shown wearing a white mask completely obscuring her scarred face.

In 1939, a couple watches a newsreel film about the exploits of The Spider-Man. The man, later revealed to be "The Magnificent Mysterio" talks to his assistant about his intentions to obtain The Spider-Man's blood, knowing that he's becoming stale from his acts, even more so after listening to a father and son choosing a show of monkeys over his act.

Meanwhile, Peter, his aunt May and Mary Jane Watson visit the New York World's Fair, discussing whether to see Salvador Dali's display or assist in Mysterio's act, choosing the latter while he reflects on the distance between him and Mary Jane. During the show, Peter's Spider Sense spikes up, but he dismisses it in order to enjoy the show. After the act, the three of them discuss how Mysterio managed to perform his escape, while Mysterio and his assistant argue about how they're wasting their time with the act. Mysterio says that once he gets The Spider-Man's blood, he will become the new Kingpin of Crime, only to be interrupted by his associate, Wilson Fisk, the actual Kingpin of Crime himself who announces him that he got someone who he can talk to.

They both arrive to a secret hideout where the Kingpin holds The Ox captive. Mysterio asks him about the Spider-Totem that he assures endowed The Spider-Man with his powers but he reveals that all of the spiders in there have died. Then he asks him about The Spider-Man's former lover Felicia Hardy. Once Mysterio and the Kingpin arrive, they question her about his whereabouts. Kingpin shoots her butler in order to intimidate her, but she shocks them once she reveals her disfigured face, however, Mysterio decides that The Spider-Man will come to them now that they got Felicia on their hands.

Afterwards, Mysterio announces a challenge to The Spider-Man requesting a Blood Sacrifice in order to save Felicia, and if he doesn't show up, she will be the one who will be sacrificed, prompting Peter to retake the mantle of The Spider-Man. Once the day arrives, Peter shows up, not willing to take his chances against Mysterio should he dare to kill Felicia in public. Mysterio sets up the trap against him, knocking him down with a sleeper gas in order to contain him. Once he's put in a special chamber, Mysterio's assistant extracts a sample of his blood in a vial, then Mysterio announces a "challenge" to accomplish his feat of escaping the water filled chamber, a plan to kill him while blaming faulty equipment.

Peter manages to improvise an oxygen mask with his webbing, giving him enough time to regain his strength and break the chamber. He frees Felicia while he taunts Mysterio by announcing that due to the lack of a sacrifice, Mysterio will be paying refunds to all the assistants. They both fight until they're suddenly interrupted by a flash of light bringing Karn, the mysterious totemic predator who proclaims that The Spider-Man is a weak spider worth the killing. Peter fights him off but then the Superior Spider-Man arrives in time, intercepting the battle in order to retrieve The Spider-Man, escaping back through his portal. Karn also escapes, vowing to find and kill all the spiders, while Felicia takes advantage of the confusion in order to steal the vial with Peter's blood.

Afterwards, both Spider-Men arrive in the year 2099 where the Superior Spider-Man brings The Spider-Man up to date with his whereabouts and the hunt of all the spiders across the multiverse, bringing him to his gathering of spider powered beings.
